Character Profile
Derek isn't much of a leader. He's stubborn, old-fashioned, and can't communicate his way out of a paper sack. The worst part is that, when you boil it down to the dregs, Derek doesn't want to be the leader. He's been alone awhile, and always a follower before that. He's the little brother strapping on boots into which he has yet to grow.
But now he's Alpha, and that responsibility can't be renounced or ignored. Derek knows from experience that all it takes is one weak link to destroy a pack, and if being cruel is the way to make his pack strong, he'll do it. It's hard to be aloof and distant; Derek grew up surrounded by a big family and seeks that connection with his pack, but has no freaking clue how to balance authority with intimacy. The end result is something of a benevolent tyrant, cards kept close to the chest, dishing out tough love, all in a misguided attempt at protecting others.

Compared to the teen cast of the show, Derek is constructed as a mysterious older guy type and less is known about his upbringing. It's known explicitly that Derek, unlike any of the teen cast, was born a werewolf to a family of mixed wolf and human origin. Derek was "at school" when his family was murdered in their home by an arsonist "six years ago." (It's unclear whether this means he was in class at high school or away attending college as his age is not stated.) By the end of season 1, it's revealed that Derek's romantic affair with Kate (member of the Argent hunter family) led to the attack on the Hale family.
After the death of his family, Derek left Beacon Hills for New York, presumably where he stayed until a few days before the series begins. He returned to Beacon Hills because his remaining family member, an elder sister named Laura and the de facto Alpha of their demolished pack, had gone missing. Upon finding her dead, Derek resumed living in the remains of the Hale house while searching for the newly arisen Alpha.
At the culmination of the first season, we learn Derek and Laura's comatose uncle, Peter Hale, had awoken deranged and hell bent on revenge. He fatally battled Laura for the position of Alpha, and Derek must in turn kill him to protect the people Peter has targeted. Derek's role in season 2 is as Alpha, and he begins building a new pack from the student body of Beacon Hills High School.
Unique Attributes
Werewolves in Teen Wolf canon are seen in multiple forms: Laura's corpse was first seen as indistinguishable from a typical wolf; Peter Hale is shown transformed in a fully-furred, monstrously muscled shape several times, his body primarily bipedal but also snouted and able to run quadruped; and most commonly, a humanoid demi-wolf form distinguished by the growth of sharp teeth, claws, facial fur, elongated ears and a pronounced brow ridge.
These physical changes are accompanied by great strength, speed, stamina, sensory ability, capacity for healing, and more. Alpha werewolves benefit from an even greater level of superhuman power, and it increases the larger their pack becomes or the more pack members they have around them. Alpha werewolves are the most likely to produce a new wolf when biting a human. A stranded, abandoned, or lone wolf is the weakest type of werewolf.
In addition to the poisonous effects of Wolfsbane/aconite, werewolves are susceptible to deadly side effects when inhaling it, touching or consuming it, or being wounded by a weapon containing it. The "Mountain Ash" seen in series is most likely the ashen remains of Rowan trees. It has been shown to repel or contain werewolves, or make the human body inhospitable to the werewolf "virus."
To make Derek more vulnerable, he should have his ability to heal reduced or slowed. He will not arrive at full strength due to the absence of most of his pack, but in the event that his pack grows, his ability to transform into the more powerful Alpha form should be capped. As Alpha, he has the ability to turn humans into werewolves. Use mod discretion in deciding whether or not he can retain this; if he does, a permissions post will be written.

Suitability
Teen Wolf is an action/comedy/thriller/horror/romance/mystery/all of the above teen drama that is notably dark in its themes, characters and setting. There are several instances in the show where scenes of graphic gore and other disturbing elements are shown. Derek's character is personally involved in many such instances.
In summation, he's a friggen werewolf.
